Summary

 

Dengue fever is a febrile disease caused by dengue virus infection, mainly manifested as fever, headache, muscle and joint pain, and rash. Most of the infection manifests as dengue fever, and may also develop into severe clinical manifestations and life threatening dengue hemorrhagic fever and dengue shock syndrome. Generally speaking, at the time of initial infection, IgM appears in the patient's blood 3 to 5 days after the onset, while IgG can only be detected 5 to 7 days after the onset, and the detection rate gradually increases. IgM antibodies not only appear earlier than IgG antibodies, but also rise faster. On the 8th day of the onset, the detection of IgM reached its peak. The IgG antibody reached the peak of detection on the 15th day of the onset.Because the IgM antibody in patients with secondary infection develops late, and even some patients may not produce IgM antibody response, the combination of IgM and IgG antibody detection, is helpful in determining the status of dengue infection.

Dengue fever infection detection methods include virus isolation method, agglutination inhibition test, virus nucleic acid detection technology, dengue NS1 antigen detection technology, and dengue specific antibody detection technology. Dengue-specific antibody detection techniques include enzyme-linked immunosorbent assay, immunofluorescence assay, rapid immunochromatography, dot immunogold test. The early virus isolation method and virus nucleic acid detection technology were cumbersome to operate, took a long time, and required high requirements for laboratories and experimenters. The joint detection of dengue-specific antibody IgG and IgM is sensitive, rapid and easy to operate. IgM antibodies appear 3-5 days after infection, IgG antibodies appear 5-7 days after infection, patients with secondary infection may not appear IgM antibodies. Early diagnosis of dengue plays a key role in controlling the epidemic.

As the content above indicated that the specific antibody against Dengue virus could be a marker to aid in the diagnosis of Dengue fever infection. In addition, no literatures showed that the dengue fever infection and detection will be influenced by the ethnic difference.

The Dengue Rapid Test Cassette (Whole Blood/Serum/Plasma) is a rapid test that utilizes a combination of Dengue antigen conjugated with colloid gold particles for the detection of IgG and IgM Dengue antibodies in human whole blood, serum, or plasma.

 

Convenient Procedure

1. Bring the pouch to room temperature before opening. Remove the test cassette from the sealed pouch and use it within one hour. If the relative humidity is greater than 60%, the test should be performed as soon as the pouch is unsealed.

2. Place the test cassette on a clean and level surface.

• For Serum or Plasma Specimens:

Ø To use a dropper: Hold the dropper vertically, draw the specimen up to the Fill Line (approximately 5 μl), and transfer the specimen to the specimen well (S) of the test cassette, then add 1 drop of buffer (approximately 40µl) to the buffer well (B) and start the timer. Avoid trapping air bubbles in the specimen well.

• For Whole Blood (Venipuncture) Specimens:

Ø To use a dropper: Hold the dropper vertically, and transfer 1 drop of whole blood to the specimen well (S) of the test cassette(approximately 14 μl for 1 drop of whole blood, draw the specimen about 1cm above the Fill Line to ensure enough sample can be transfer), then add 1 drop of buffer (approximately 40 µl) to the buffer well (B) and start the timer.

3. Wait for the colored line(s) to appear. The test result should be read at 10 minutes.

Do not interpret the result after 20 minutes.
